The US rules say that once per round you may put one card FROM YOUR HAND face down under the museum.
In a game here I destroyed a museum which had 4-5 cards underneath. That player suddenly lost 5 built districts! Does this mean that we must put a built card (from our city) underneath the museum, and not a card from our hand??

The rule of this card is as you have said in the first paragraph. When you destroy a Museum with 5 cards underneath, that player loose the points of thge Museum (4 points) plus the 5 points that the Museum granted him.
No anyone district are destroyed. I cannot believe that such a think have occur.
